🤨 质 (substance) + 疑 (doubt) = doubt the substance of a claim = challenge it
质 carries the sense of quality or substance, and 疑 means doubt or suspicion. Put them together and you get probing the substance of what someone said, which is exactly what it means to call a claim into question.
